Research Overview

Kisspeptin-10 Research Standard

Kisspeptin-10 (CAS 374675-21-5) is the decapeptide C-terminal fragment of the KISS1 gene product, supplied as a 10 mg lyophilized powder in a 3 mL vial. It sits at the top of the hypothalamic-pituitary-gonadal axis: kisspeptin acts on GnRH neurons, GnRH acts on the pituitary, and the pituitary acts on the gonads. Working one step upstream of GnRH is what makes it a distinct tool rather than a redundant one.[1]

Mechanism of Action & Research Context

The molecular target is KISS1R, also known as GPR54, a G-protein-coupled receptor expressed on GnRH neurons.

  • An obligatory upstream signal. Kisspeptin binds KISS1R on GnRH neurons and stimulates their activity, which in turn provides an obligatory signal for GnRH secretion. The word matters: this is not a modulatory input that fine-tunes the axis, it is a required one, and removing it gates everything downstream.[1]

  • Integration point for multiple inputs. Kisspeptin signalling is itself the target of steroid hormone feedback and of nutritional and metabolic regulation — which is why it is studied as the node where metabolic state reaches the reproductive axis.[1]

  • Antagonists exist and are useful controls. Four peptide antagonists and one small molecule antagonist have been designed for this receptor. An agonist experiment without an antagonist arm leaves receptor dependence untested.[1]

  • The neurons it acts on have an unusual history. GnRH-1 neurons originate in the olfactory placode and vomeronasal organ, then migrate along vomeronasal nerves through the cribriform plate into the hypothalamus. Failure of that migration causes failure of the reproductive axis, sometimes alongside anosmia — the developmental link behind Kallmann syndrome.[2]

Research Applications

Primary fields of in vitro and preclinical laboratory investigation include:

  • KISS1R binding and functional assays with peptide or small-molecule antagonist controls.[1]

  • GnRH neuron activity measured by electrophysiology or secretion readout.[1]

  • Metabolic and steroid feedback studies at the kisspeptin node.[1]

  • Neuronal migration and development models.[2]
